With a lack of villains in the current timeline/season of MLP, it's safe to say that an solid antagonist is needed. Who would be able to take up that kind of thing? Well, why not the character that was parasitically based on the single most famous antagonist/super villain in history? Well the Joker of course, and with Mane-iac from the episode "Power-ponies", it seems fitting that she is the next in line to bring anarchy/chaos to the peaceful lands of Equestria. This will be written/told through separate tales/short stories that are based around popular Joker/Batman comics, such as "The Killing Joke", "Death in the Family", etc.
